Poly-L-Lysine solution is intended for use as an adhesive subbing solution for immunohistochemistry(IHC) and routine histologic staining preparations. The loss of paraffin and frozen sections from slides has long been a problem during routine Histologic staining procedures. Various adhesives including albumin, gelatin and chrome alum have been applied to slides to minimize this loss. Different solutions of Poly-L-Lysine have been shown to be most effective in promoting adhesive of sections. The polycationic nature of this molecule allows interaction with the anionic sites of tissue sections resulting in strong adhesive properties. Poly-L-Lysine has been demonstrated as an effective tissue adhesive for use in various microwave procedures. 簡介 多聚賴氨酸溶液是廣泛應用的組織切片與玻片黏合劑，該多聚陽離子分子與組織切片上的陰離子相互作用會產生較強的黏合力。 適用組織學，免疫組織化學，冰凍切片，細胞涂片，原位雜交等使用的玻片的防脫片處理，以防實驗操作過程中組織掉片。也可用于細胞培養，增加細胞貼壁能力。 Poly-L-lysine的中文名為多聚賴氨酸，簡稱PLL。本Poly-L-lysine為Poly-L-lysine hydrobromide，分子式為L-Lys-(L-Lys)n-L-Lys·xHBr，分子量為150,000-300,000，CAS Number 25988-63-0。 本產品為進口分裝。分子量大于70,000的多聚賴氨酸可以用于促進細胞貼壁生長，本產品可以用于促進細胞的貼壁生長。本產品也可以用于組織學(Histology)分析時的粘片劑。 使用說明： 1． 滅菌的蒸餾水1:10稀釋該多聚賴氨酸溶液。 2． 用之前將稀釋的多聚賴氨酸溶液放在室內，使其溫度到室溫18-26℃ 3． 將玻片浸在稀釋的多聚賴氨酸溶液5分鐘。注意增加時間不會提高包被效果。 4． 在60°C烘箱1小時干燥，或室溫18-26℃過夜干燥待用。 注意事項： 1． 每100ml已稀釋的多聚賴氨酸溶液要包被的玻片40-90張，超過90張片子將影響其黏合力。 2． 用之前的玻片必須保持清潔。必要時用含1% HCl的70%乙醇溶液來清洗。 3． 不要在用過的稀釋液中加新的溶液。 4． 釋過的多聚賴氨酸溶液要放在2-8℃，至少在3個月內是穩定的。 5． 用過的稀釋液要過濾，若出現渾濁或長菌要丟棄。 |
